PURPOSE:DNA coding polypeptide having G-CSF activity, obtained by the use of a transforming host from mRNA prepared from a mouse cell having granulocyte colony stimulation factor (G-CSF) activity. CONSTITUTION:mRNA of NFSA strain of mouse fibrosarcoma is treated with a reverse transcriptase by using oligo (dT) as a primer to give duplex cDNA, an EcoRI linker is added to the cDNA and the duplex cDNA is linked to lambdagt10 vector to prepare cDNA library. Plaque hybridization is carried out by using DNA fragment containing human G-CSF cDNA from the library to give one hybridizing clone. EcoRI fragment of the clone is subcloned with a cloning vector to give the aimed mouse G-CSF cDNA. |
