METHOD FOR MEASURING LIPID PEROXIDE

PURPOSE:To perform simple and exact colorimetric measurement of the quantity of liquid peroxide by migrating the lipid peroxide in a sample into an org. solvent by extraction and bringing the lipid peroxide and color former into reaction in the presence of the modified heme protein in the org. solvent. CONSTITUTION:The extraction of the lipid is quantitatively executed in a manner as to avoid the intrusion of the non-protein component such as free sugar and amino acid. The color former which is oxidized by the lipid peroxide in the presence of the modified heme protein and is colored by the same is usable and the kind thereof is not particularly limited. The modified heme protein is the heme protein which is made to have solubility in the org. solvent while the effect of accelerating the oxidation reaction is maintained. The reaction is effected by mixing the three; the org. solvent contg. the lipid peroxide, the color former dissolved in the org. solvent and the modified heme protein dissolved in the org. solvent and incubating the mixture under the prescribed conditions. The degree of coloration is determined by measuring the absorbancy. The quantity of the lipid peroxide is determined by comparing the determined absorbancy and the absorbancy determined in the same manner with the standard lipid peroxide.
