<p>PURPOSE:To obtain an antibody applicable to immunoassay capable of simply, easily, sensitively and quickly measuring trypsin by culturing hybridoma for producing the specific monoclonal antibody of tripsin and trypsinogen and extracting an antitrypsin monoclonal antibody from a cultured substance. CONSTITUTION:To obtain hybridoma to which antitrypsin antibody producing lymphocyte and a myeloma cell are fused, trypsin is sensitized to a mammal (mouse or the like). The lymphocyte is extracted from an immunized animal. An antigen refined from human pancreatic juice or the like is used. It is favorable to use the myeloma cell originating in the same species as the immunized animal. Then, after suspended and mixed in a culture medium such as MEM or the like, the lymphocyte and the myeloma cell are fused by using a fusion accelerator. After washed and removed with the fusion accelerator, only the hybridoma is raised by culture. The hybridoma is cultured by using the culture medium and an antitrypsin monoclonal antibody is extracted from a supernatant liquid.</p> |
