MODIFIED HUMAN TISSUE-TYPE PLASMINOGEN ACTIVATOR AND ITS PREPARATION

Human tissue-type plasminogen activators (t-PA) are characterised by having a modified or no carbohydrate structure at amino acid residue 117. Such t-PAs have substantially the same activity and binding characteristics as native t-PA but have an increased in vivo half life. They may be prepared by site mutagenisis corresponding to amino acid residue positions 117 or 118 or 119 or by enzymic removal of most of the carbohydrate at position 117 or native t-PA using an endoglycosidase. The carbohydrate structure at amino acid residue positions 184 and 448 remains unmodified.
