<p>PURPOSE:To remarkably enhance protoplast formation ratio of an arboreal plant and impart high biological activity, by treating leaves of a cut arboreal plant with an enzyme containing pectinase, hemicellulase and cellulase. CONSTITUTION:Leaves of an arboreal plant, e.g. oleander, apple, hydrangea, nandin, etc., are washed with water, sterilized, washed with sterilized distilled water and aseptically cut. The cut pieces of the leaves are then washed with a washing solution, e.g. protoplast washing solution, etc., and an enzymic solution containing pectinase, hemicellulase and cellulase is then added to carry out enzymic reaction at about 25-35 deg.C for several hr. After completing the reaction, filtration is carried out an the filtrate is centrifuged at a low speed to give a precipirate fraction. The precipirate fraction is washed with the protoplast washing solution 2-3 times to produce a crude protoplast solution. The crude protoplast provides normally >=60% protoplaslt formation ratio.</p> |
