<p>PURPOSE:To improve a substance-productivity of a recombinant, by subjecting E.coli to simultaneous infection with a specific dC-type recombinant T4 phage and a specific surviving T4 phage, thereby effecting DNA recombination between both phages in the infected bacterial cell. CONSTITUTION:A dC-type recombinant T4 phage containing an inserted DNA fragment is prepared beforehand. The DNA fragment is effective in coding a genetic information to be expressed in a T4 phage nonessential gene other than gammaII gene and is inserted in a state to enable the expression on a phage genom. An E.coli cell is subjected to simultaneous infection with the above recombinant T4 phage and a surviving T4 phage having normal gammaII gene and containing hydroxymethylcytosine in the DNA to effect the DNA recombination between both phages in the infected bacterial cell. A surviving recombinant T4 phage produced by the above process contains an inserted DNA fragment coding a genetic information to be expressed, has normal gammaII gene and contains hydroxymethylcytosine in the DNA.</p> |
