PURPOSE:To obtain a plasmid having a DNA sequence coding industrially useful lipase, e.g. 1,3-regiospecific lipase, thermostable lipase, etc., and produce the lipase using the plasmid, by determining the DNA sequence. CONSTITUTION:A DNA sequence capable of cloning a 1,3-regiospecific lipase gene held by Pseudomonas fragi which is one of the genus Pseudomonas and coding lipase is determined or DNA sequence capable of cloning heat-resistant lipase gene held by Bacillus stearothermophilus which is one of the genus Bacillus and coding the lipase is determined to carry out comparison with well-known lipase and investigation. As a result, the DNA sequence corresponding to the amino sequence expressed by the formula (A, X, Y and B are amino acid) is found to be DNA sequence common to the lipase. A plasmid having the above-mentioned DNA sequence, e.g. DNA sequence capable of coding lipase of the Pseudomonas fragi expressed by the formula (A is Ileu; X is His; Y is Gln and B is Ala), is used to produce the aimed lipase. |
