发明名称 NOVEL COSMID VECTOR, DERIVED FROM LAMBDA PHAGE AND HAVING REPLICATION FUNCTION AND PRODUCTION OF GENETIC BANK USING SAID COSMID VECTOR
摘要 PURPOSE:To readily and efficiently produce a genetic bank of an extraneous DNA using a novel cosmid vector, by preparing the novel cosmid vector characterized by having replication function derived from lambda phage employing a genetic manipulation technique. CONSTITUTION:A cosmid vector having >=2 COS of lambda phages or lambdoid phage with the same orientation property in the same molecule and replication function derived from the lambda phage is cleaved with 2 kinds of restriction enzymes to give the right arm and left arm of almost the same length each having at least one COS and various kinds of extraneous DNA fragments having 35-45kb base pairs are inserted between the arms to convert the arms into tranducing phage particles by packaging. The resultant particles are then cloned in Escherichia coli to produce a genetic bank of the extraneous DNA. Since the number of copies of recombinant DNA per cell in a host is large and almost constant, the quality and stability of the genetic bank are high.
