TESTING METHOD FOR ENZYME ACTIVITY

PURPOSE:To make the quick and easy non-destructive test of the enzyme activity of an immobilized enzyme film with high accuracy by irradiating light of prescribed wavelength to the surface of the immobilized enzyme film and measuring the absorbancy thereof. CONSTITUTION:An O-ring fixing part 31 for holding the immobilized enzyme film is provided to one side of an immobilized enzyme film holder 30. Said fixing part with the film side thereof faced to the outside is fitted into an O-ring 32 and is fixed to the fixing part 31. A buffer soln. injecting port 33 is provided to the other side of the holder 3. The irradiation of the light of the prescribed wavelength to the surface of the immobilized enzyme film is executed only in the part A brought in contact with an indicating electrode surface by the holder 30. The quantity of the enzyme in the immobilized enzyme film is thus evaluated by measuring the absorbancy of the light of the prescribed wavelength without inducing an enzyme reaction in the above-mentioned manner and therefore, the time for evaluation is made shorter and the operation is made simpler. The non-destructive test is thus made possible. Since the part to be irradiated with the light is limited to the film surface in contact with the indicating electrode and therefore, the correlation between the absorbancy and the enzyme electrode is improved and the accuracy of the evaluation is improved as well.
