PRODUCTION OF COLONY STIMULATION FACTOR (CSF)

PURPOSE:To enable the mass-production of CSF, by establishing CSF-producing cell strain able to be subcultured over a long period from a gastric tumor tissue exhibiting neutrophilia free from symptom of hypercalcemia and culturing said cell strain. CONSTITUTION:Tumor cell is extracted from a gastric carcinoma patient exhibiting neutrophilia free from symptom of hypercalcemia and transplanted to a nude mouse. The cell is subcultured for several generations and a strain exhibiting leukocytosis in peripheral blood of nude mouse is selected. A thin section of the tumor tissue is subjected to monolayer cell culture by dispase treatment and coexisting nude mouse-originated cells are removed by the use of a rabbit antiserum against fibroblast cell originated from cultured nude mouse spleen. The strain is subcultured for 12 generations and cloned by limiting dilution method to establish a CSF-producing cell strain. The cell strain is cultured in a serum-containing medium until the system reaches a confluent state, when the medium is exchanged to a serum-free medium. CSF is produced by culturing the strain in the serum-free medium.
