PURPOSE:To obtain a mouse monoclonal antibody from a mouse and use the antibody for diagnosis, etc., by transforming peripheral blood lymphocytes of a human of convalescent hepatitis non A non B, etc., with an EB virus and using the resultant monoclonal antibody. CONSTITUTION:The peripheral blood of human or chimpanzee of convalescent hepatitis non A and non B is subjected to heparinization and collected and lymphocytes are separated and brought into contact with an EB virus solution and cultivated. The cultivation supernatant is collected to carry out screening of antibody activity using hepatic slices with onset of hepatitis non A and non B. Cloning is then carried out for cultivated cells positive to antibody activity to obtain an antibody produced by the above-mentioned cloned cellular strain. A mouse is immunized with an antigen appearing in the hepatic cells with the onset of hepatitis and splenic cells thereof and mouse myelomatous cells are subjected to cell fusion to select a hybridoma and give cells positive to the antibody, which are then cloned to afford there aimed antibody. |
