PURPOSE:To obtain a novel acyl-CoA synthetase, having improved preservation stability and capable if exhibiting sufficient activity even a temperature normally used in tests, by cultivating a thermophilic microorganism belonging to the genus Pseudomonas and collecting the synthetase from the culture. CONSTITUTION:A thermophilic microorganism which is Pseudomonas UKSW-3733 strain having 45-55 deg.C optimum growth temperature is cultivated in a liquid culture medium containing an organic acid, alcohol, amino acid, inorganic salt, etc., at 35-60 deg.C under aerobic condition. The aimed acyl-CoA synthetase is isolated and purified from the resultant microbial cells using crushing, extracting, salting out, column chromatography, etc. The above-mentioned synthetase is capable of acting on a saturated or unsaturated lower - higher fatty acid in the presence of adenosine triphosphate and coenzyme A to produce a CoA derivative of the fatty acid, adenosine monophosphate and pyrophosphoric acid. The optimum working temperature is 50-60 deg.C. |
