PURPOSE:To quantitatively analyze calcium ions in an aq. liquid sample by providing a reagent layer contg. at least one kind of indicator which can make an optically detectable change by conjugating with the calcium ions onto a light-transmissive water-impermeable base. CONSTITUTION:A protein-impermeable, hydrophilic nonporous intermediate layer is provided between a porous developing layer and reagent layer of the monolithic type multi-layered analyzing element for quantitative determination of the calcium ions. A pH buffer is incorporated into at least either of said layers. The diffusion and transfer of serum protein such as albumin to the reagent layer is prohibited and the diffusion and transfer of the protein- conjugated calcium to the reagent layer are substantially prohibited, by which the calcium ions which can be diffused and transferred thereto are detected by the reagent layer. The quantitative analysis of the calcium ions is thereby executed without an independent fractional operation; in addition, the quantitative analysis of the calcium ions is executed without diluting the aq. body fluid sample or at <=10 times dilution ratio.
