PURPOSE:To provide a method for determining a specific substance in a specimen characterized in that hydrogen peroxide existing in a specimen or produced by a reaction is eliminated by reacting with phenol or aniline, etc., in the presence of a peroxidase. CONSTITUTION:Hydrogen peroxide existing in a specimen or produced by the reaction of an oxidase with a specimen containing a substance oxidizable with oxidase to produce hydrogen peroxide is made to react with a phenolic compound, etc., of formula in the presence of a peroxidase. The hydrogen peroxide is decomposed by this process and the substance to be determined is converted to a substance oxidizable with an oxidase by enzymatic reaction. The produced substance is oxidized in the presence of an oxidase to produce hydrogen peroxide. The objective substance can be determined by determining the produced hydrogen peroxide.
