The invention relates to a reagent and a method for detecting, in a biological medium, an element capable of acting in a ligand/affinity agent system. The reagent consists of a suspension of particles supporting the counter-element of the ligand/affinity agent system, within a solution coloured by a first colorant, the said particles being coloured with a different counter-colorant, so that the suspension has an apparent monochromatic coloration distinct from those of the above indicated colorant and counter-colorant. Agglutination of the particles, by virtue of the presence of the element contained in the biological medium is manifested by the appearance of a double coloration within the reagent.
