PRODUCTION OF PROTEIN BY EXTRACELLULAR SECRETION

PURPOSE:To efficiently obtain a desired protein in a large amount using a strain, e.g. Escherichia coli, as a host, by integrating a gene coding an exogenous protein in a vector multiplicable in a host microbial cell to transform the host and specifically adjusting the multiplication process thereof. CONSTITUTION:A vector having a gene capable of coding a promoter and a gene capable of coding a signal sequence under the control thereof and multiplicable in a predetermined host microbial cell is prepared. A gene capable of coding an exogenous protein is integrated into the above-mentioned vector to prepare a recombinant DNA, which is then integrated into a host microbial cell to transform the microbial cell. The resultant transformant is cultivated in a culture medium containing a promoter activity regulating substance in an amount required for causing induction of protein synthesis ability from the latter period of the logarithmic growth phase to the first period of the resting phase in the multiplication process of the microorganism and then subjected to cultivation by adding the promoter activating substance from the outside. The resultant exogenous protein is recovered from the culture fluid.
