STABILIZED HUMAN PROUROKINASE

摘要

PURPOSE:To improve stability of human prourokinase to enzyme in and out of living body, by forming a human prourokinase-like polypeptide having an amino acid sequence represented by a specific general formula or an amino acid sequence essentially same as the above sequence. CONSTITUTION:A DNA segment coding a polypeptide of formula (Met is methionine; Ser is serine at the 1-site of N-terminal; X is lysine or amino acid at 135-site other than basic amino acid; Y is phenylalanine or acidic amino acid at 157-site; horizontal line is amino acid sequence same as the corresponding part of the amino acid sequence of natural human prourolinase) is prepared beforehand. Escherichia coli transformed with said DNA segment and plasmid is cultured and the objective prourokinase-like polypeptide is separated from cultured bacterial cell.
