<p>PURPOSE:To obtain the aimed elastase, by cloning a human pancreatic elastase, ingressing the resultant recombinant DNA molecules into a host and expressing the pancreatic elastase gene. CONSTITUTION:An mRNA is separated from the human pancreas and used as a template to synthesize a complementary double stranded DNA. The resultant recombinant plasmid is then introduced into to transform Escherichia coli (RRI strain, etc.). Rat elastase IIcDNA is used as a probe in the resultant recombinant to determine a clone having DNA completely cloning a human pancreatic elastase expressed by the formula. Elastase gene is cut out of the resultant clone and linked to a suitable expression vector and introduced into a suitable host for expression. Thereby, the aimed elastase having the biological activity equal to that of the elastase extracted and purified from human pancreatic juice is obtained.</p> |
