PURPOSE:The titled DNA suitable for industrial use of gene recombination, obtained by integrating vector DNA for other organism into cyclic duplex DNA having a specific base sequence. CONSTITUTION:Mungbean Yellow Mosaic Virus (MYMV) is purified and extracted to give single stranded DNA having length of 2675 bases. Then the single stranded DNA is reacted with a primer derived from calf thymus gland and a duplex enzyme such as reverse transcriptase of AMV, etc., to give cyclic duplex DNA consisting of base sequence shown by the formula. Then this duplex DNA is decomposed with a restriction enzyme (e.g., BamHI) to give a decomposition solution, which is blended with a BamHI decomposition solution of vector DNA (e.g., plasmid pBR322 for Escherichia coli) for other organisms and bonded and subjected to ring formation with a DNA bond enzyme to give hybrid DNA. |
