The photometric determination of protein C, especially in plasma, entails the sample containing the protein C to be determined being incubated with thrombin with the formation of activated protein C, then excess thrombin inhibitor such as antithrombin III being added and subsequently the decrease in the formation, mediated by coagulation factors, of thrombin from prothrombin being determined by means of a chromogenic thrombin substrate such as H-D-Phe-Pip-Arg-pNA or Tos-Gly-Pro-Arg-pNA. <IMAGE> |
