NEW MATERIAL:A human immunoglobulin that binds with a stomach cancer cell strains, MKN-1, MKN-45 and KATO-III, lung cancer cell strains, PC-10 and PC-14, but does not to hematopoietic cell strains, CCRF-CEM and U-937. USE:Diagnostic agent and remedy for stomach and lung cancers. PREPARATION:Lymph nod cells are collected by extracting lymph nods near the area affected by stomach cancer in a patient surgerical operation and by fragmenting the nods. After proliferation, the cells are combined with the B-cell strain, ATCC.CRL-8118 and poured into individual wells on microplates and cultured. Then, the wells which have formed colonies are transferred to cultivation plates to effect propagation. Further, the supernatant of the cultivation mixture is added to each well on a microplate into which the target cells are poured to effect the reaction. Then, the strains reactive with the cancer cells are selected and cloned and cultivated to obtain a human imminoglobulin reactive with cancer cells. |
