PURPOSE:To efficiently purify RNA useful for genetic engineering method, by bringing RNA obtained from a polysome franction into contact with glyceryl porous glass containing oligothymidylic acid to adsorb mRNA, separating rRNA and eluting mRNA. CONSTITUTION:Precipitate of a solution of eruthrocyte is dissolved in guanidine thiocyanate to give a solution, to which an alcohol is added to give precipitate, the precipitate is dissolved in an aqueous solution of guanidine hydrochloride to give a solution, further ethanol is added to the solution to give precipitate, the precipitate is collected by centrifuging and dissolved in a buffer solution to give a polysome RNA solution containing mRNA. Then, it is brought into contact with glyceryl porous glass containing oligothymidylic acid to adsorb mRNA, rRNA is separated and MRNA is eluted with a tris-hydrochloric acid buffer solution, to purify mRNA. |
