PURPOSE:To enable analysis with high sensitivity by adding successively specific three kinds of reagents to a sample contg. catechol amine, converting the catechol amine to trihydroxy indol (THI), extracting THI in a solid phase and subjecting the same to high speed chromatography. CONSTITUTION:Catechol amine is separated and extracted from a sample contg. catechol amine in blood plasma, etc. A potassium phosphate buffer contg. potassium ferricyanide, aq. ascorbic acid soln. and NaOH soln. are added in this order to the extraction liquid and thereafter the reaction liquid is adjusted to 3-5pH and is subjected to reversed phase chromatography of a silica-packed column to adsorb the formed THI. The eluted THI soln. is then subjected to high speed chromatography for fluorometric analysis. The chart having the sharp peaks of the respective catechol amines without spreading and cracking is thus obtd. and the analysis with high sensitivity and good reproducibility is made possible. |
