<p>PURPOSE:The titled antibody, obtained by administering a specific hybridoma to a mouse, etc., converting the hybridoma into ascitic cancer, collecting a monoclonal antibody from the abdominal dropsy and recognizing a sialated glycoprotein as an antigen, having improved reaction to pulmonary adenocarcinoma and useful for blood serum diagnosis of pulmonary cancer. CONSTITUTION:Mouse splenic cells immunized and prepared by using 2-5X10<6> cells/head of tissue cells of human pulmonary cancer obtained from immunologically tolerated mouse of 20-400mug/head of tissue membrane component are mixed with 8-azaguanine-resistant mouse myelomatous cells, etc., at 5-10:1 ratio and subjected to fusion and cultivation to give a hybridoma strain ALC-186, etc., having specificity for human pulmonary adenocarcinoma and capable of producing monoclonal antibodies. 2-4X10<6> cells/head resultant hybridoma cells are then injected into the abdominal cavity of a mouse to form ascitic cancer. A purified monoclonal antibody is then separated from the abdominal propsy and subjected to antigenic analysis to afford the aimed antihuman pulmonary adenocarcinoma specific monoclonal antibody capable of recognizing sialated glycoprotein or glycolipid as an antigen.</p> |
