PURPOSE:A shuttle vector, containing an autoreplication sequence of Candida maltosa, Leu2 gene derived from Saccharomyces cerevisiae, ampicillin-resistant gene and tetracyclin-resistant gene and having a wide region of hosts. CONSTITUTION:A gene library of Candida maltosa is prepared by using a vector YEp13 of Saccharomyces cerevisiae and transformed by using Candida maltosa having requirement for leucine as a host. A plasmid is separated from the resultant transformant to transform Escherichia coli. An ampicillin-resistant plasmid is then separated to isolate a region (TRA region) required for transforming Candida maltosa with a restriction enzyme BamHI. The vector YEp13 is cleaved by a restriction enzyme BglII and a vector pBR322 of Escherichia coli is cleaved by a restriction enzyme EcoRI. Both are then linked and the resultant linked plasmid is cleaved by a restriction enzyme XhoI and linked to the TRA region. |
