<p>PURPOSE:To obtain a monoclonal antibody effective for chemical-resistant cancerous cells with slight side effects, by using a hybridoma obtained by fusing splenic cells prepared by immunizing a mouse with a specific chemical-resistant microorganism to mouse myelomatous cells. CONSTITUTION:A mouse is immunized with an adriamycin(ADM)-resistant strain K562/ADM selected from human myelogenic leukemic cell strain K562 to give immunize splenic cells, which are then fused to mouse myelomatous cells to afford fused cells. The resultant fused cells are cultivated in a selective culture medium to prepare a hybridoma capable of producing an antibody. The resultant hybridoma is selected and cloned. The clone is then cultivated in a culture medium, etc., to produce the aimed monoclonal antibody, having the ability to specifically discriminate the ADM-resistant strain, inhibiting the cell multiplication of the ADM-resistant strain or enhancing the sensitivity of the cells to vincristine or actinomycin D and belonging to the IgG isotype in the cultivation supernatant liquid.</p> |
