PURPOSE:To expand the upper limit of a measuring concentration range of an enzyme electrode, by using a relationship curve between the reciprocal of a substrate concentration and the reciprocal of an output of the enzyme electrode as calibration curve in the assay of a substrate. CONSTITUTION:An immobilized alcohol oxidase film is mounted on a Clark type hydrogen peroxide electrode to form an enzyme electrode and a ethanol reference solution is brought into contact with the electrode to obtain a relationship between the ethanol concentration and the reaction output thereof. Then, a relationship is determined between the reciprocal of the substrate (ethanol) concentration and the reciprocal of the reaction output (Lineweaver-Burk plot) and the assay of the substrate concentration in a sample is done using the resulting linear line as calibration curve. |
