PURPOSE:To produce the desired HBsAg and HBsAg attached with pre-S, in high yield, by culturing a specific transformed yeast cell in the presence of galactose preventing the suppression of galactokinase promotor. CONSTITUTION:A plasmid vector containing a gene to proliferate the recombinant plasmid in yeast, a gene which is a selection marker to release transformed yeast cell strain by the introduction of a recombinant plasmid, and a gene of a galactokinase manifestation controlling region of a yeast cell is prepared beforehand. A surface antigen gene of hepatitis virus B or a recombinant plasmid integrated with a pre-surface antigen gene is introduced into the plasmid vector at a site to be controlled with galactokinase promotor, and the obtained transformed yeast cell is cultured to produce the surface antigen or surface antigen attached with pre-surface antigen of hepatitis virus B. |
