PURPOSE:A cloned maize ppc gene useful for enhancing carbon dioxide assimilation of cultured plant, etc., obtained by coding a substantially functional part of phosphoenolpyruvic carboxylase (PEPCase) of maize. CONSTITUTION:Maize poly A'RNA is propared from green leaves obtained by culture of maize on absorbent sanitary cotton. Then, it is treated with a restriction enzyme, DNA ligase, etc., to give DNA. Then, it is added to a bactotrypton medium and strain DHI of Escherichia coli is cultivated in the medium to give cDNA library. Then, the prepared strain DHI is treated with an alkali to give plasmid(pM), Escherichia coli PCR1 is transformed with the pM to give mold PCRI/pM5 and further pM52 is prepared from a single colony of this mold. Then, it is transduced to Escherichia coli K-12PCR1/pM52 and ppc is cleaved from the prepared pM to give a gene with DNA sequence comprising bases shown by the figure. |
