<p>In the 70 KDa hsp promoter, modifications brought about to the 5' and 3' non-translated sequences, including the insertion or deletion of nucleotides and the number of heat shock consensus sequence elements present in the proximal region of a heat shock gene promoter critically influences promoter activity. Sequence variants in translation initiation and 3'-non-translated regions have been shown to improve levels of induced protein production. Mutant promoters were constructed that have higher maximal activities than the natural parent promoter in heat-shocked cells. These mutant promoters are also expressed at high levels at a temperature below that required for optimal induction of the natural heat-shock genes. These results are of practical interest for attempts to use heat-shock promoters for the inducible production of eukaryotic proteins of medical or industrial importance.</p> |
