发明名称 METHOD FOR NOVEL PARTIAL STAINING OF HUMAN CHROMOSOME
摘要 PURPOSE:To enable the discrimination and positional judgement of the chromosome of a specific part difficult to discriminate heretofore, by using bis(9'- acridil)-1,4-phenyldihydrazone as a staining agent for the partial staining of a human chromosome. CONSTITUTION:Bis(9'-acridil)-1,4-phenyldihydrazone represented by formula is used as a staining agent. This acridine derivative is a novel compound and can be easily synthesized. Further, the acridine derivative is extremely unstable in the atmosphere and insoluble in water but soluble in an org. solvent such as methanol or glacial acetic acid and further takes two chemical forms by differentiating pH. That is, at pH of 3.5 or less, said derivative is present as a compound having such a structure that a proton is added to nitrogen of an acridine ring and, at pH of 6.5 or more, as a compound having a separate structure. This novel partial staining method is performed according to the technique of a fluorescent staining method except that bis(9'-acridil)-1,4- phenyldihydrazone is used.
