<p>PURPOSE:To produce a monokine derived from macrophage, by cultivating cell strain AH-01 derived from human leukemic treatment is a serum-free medium, differentiating and inducing it to a cell having macrophage-like functions and cultivating the cell in a serum-free medium. CONSTITUTION:Human bone marrow leukemic cell THP-1 is cultivated in an equine serum-containing medium with low serum, further cultivated in a medium containing no serum, to give serum-free culture cell strain AH-01. The cell is multiplied in a serum-free medium under a condition to maintain logarithmic growth of the cell until the cell concentration becomes 1-2X10<6> cells/ml, the cells in a logarithmic growth phase is recovered by centrifugal separation method, etc., the cell AH-01 is suspended in a serum-free medium containing a phorbol ester compound, mezerein, vitamin A, etc. and cultivated for 1-24hr to differentiate and induce a cell having macrophage-like functions. The cell from which a differentiating and inducing agent is removed by centrifugal separation method, etc., is suspended in a new serum-free medium and cultivated for 1-4 days to produce a monokine.</p> |
