PURPOSE:To provide a recombinant plasmid containing a gene coding meso-alpha, epsilon-diaminopimeric acid dehydrogenase (DDH), effective to impart E. coli, etc., with DDH activity and useful for the production of a large amount of DDH at a low cost. CONSTITUTION:A mixture of chromosome DNA extracted e.g. from Corynebacterium glutamicus and vector pBR322 originated from E. coli is treated with restriction enzyme EcoRI. The treated product is made to react with T4 ligase, and E. coli TM135 is transformed with the resultant reaction product. A plasmid is separated from the transformed strain, digested with several kinds of restriction enzymes, analyzed and separated by agarose gel electrophoresis, and a DNA fragments of 5.3kb and 2.7kb are inserted to the EcoRI incision site to obtain the objective recombinant plasmid pDD3 containing the gene coding DDH. |
