REAGENT FOR MEASURING RETICULOCYTE FOR FLOW SIGHTMETRY

PURPOSE:To obtain a measuring reagent increasing the fluorescence intensity of the reticulocyte in a stained specimen and lowering the background fluorescence of a specimen solution and enhanced in the measuring accuracy of the reticulocyte, by containing auramine O in a measuring reagent as the fluorescent dye of the reticulocyte. CONSTITUTION:In the measurement of a reticulocyte being an immature red corpuscle by flow sightmetry, an isotonic buffer dye solution adjusted to pH6-10 containing 20-2,500mug per ml of a reagent of auramine O along with 5-20mg of an alkali metal salt such as NaCl is used. This isotonic buffer solution is added to the liquid made anticoagulant by adding ethylenediaminetetraacetic acid to a whole blood specimen and the resulting mixture is incubated at room temp. for 10min to be sent to a fluorescent flow sight meter and irradiated with exciting light with a wavelength up to about 480nm to detect fluorescence with a wavelength of 520nm or more. By this method, intensive and strong fluorescent staining is applied to the reticulocyte without being affected by the degree of a reticulocyte ratio and the concn. of a red corpuscle to enable high accurate measurement and the reagent is utilized in the diagnosis of hemolytic anemia.
