PURPOSE:To surely develop an exogenote and to carry out genetic transformation of Coryneform bacterium, by introducing a plasmid containing a gene with a specific base sequence to a cell. CONSTITUTION:A plasmid shown by the table I which has (A) a base sequence shown by the formula I (T is thymidylic acid, M is deoxyguanylic acid of G, deoxycytidylic acid of C, or T; N is deoxyadenylic acid of A, G, T or C; P is C, or G; Q is A or C), (B) a base sequence shown by the formula II (R is A, T, or C; S is nothing or G, or A; W is T or A; T and A are as shown for the formula I) at the downstream of 15-30 base pairs of the component A, (C) a base sequence C to function as ribosome bond site in the Coryneform bacterium cell (K) at the downstream of >=7 base pairs of the component B, and (D) a base sequence to function as a translation starting codon in the cell K at the downstream of >=4 base pairs of the component C, has a gene to be developed and directly connected to the component D and produces a hydrolase, etc., as a development product, and can be replicated with the cell K is introduced to the cell II shown by the table II. |
