PURPOSE:To determine the activity of an enzyme in high accuracy and sensitivity, by condensing an aniline derivative with a phenylenediamine derivative separated from a substrate by the enzymatic reaction with the objective enzyme using bilirubin oxidase as a binder, and carrying out the colorimetric determination in the presence of an azide. CONSTITUTION:In the determination of the enzymatic activity in a biospecimen such as serum, plasma, etc., a synthetic substrate which isolates a phenylenediamine derivative by the action of the objective enzyme (e.g. peptidase) is used as a substrate. The phenylenediamine isolated by the enzymatic reaction is condensed with an aniline derivative using bilirubin oxidase as a binder. The pigment produced by the condensation reaction is determined by the colorimetric determination in the presence of an azide (e.g. sodium azide). The activity of the objective enzyme can be determined in high accuracy and sensitivity by this process.
