PURPOSE:To provide the titled DNA fragment containing a gene producing glutamate dehydrogenase (GDH) originated from a specific bacterial strain, and useful for the breeding of microorganisms to improve the productivity of amino acid or protein. CONSTITUTION:Whole DNA is extracted from the cell of coryne-type bacteria capable of producing glutamic acid, and decomposed with a vector-cleaving restriction enzyme to obtain plural DNA fragments (A) containing GDH- producing gene. A vector DNA compatible to the host strain is incised and cleaved with a restriction enzyme, and the component A is integrated to the cleaved site of the product to obtain a recombinant DNA (B). The component B is introduced into host E.coli to effect the transformation of the host. The transformation is cultured and a transformation containing a recombinant DNA having the GDH-producing gene is selected and separated. The objective DNA fragment containing the GDH-producing gene, having a molecular weight of about 5.6 Kb, containing single-chain terminals at both ends by the action of the restriction enzyme EcoRI and exhibiting the incision sensitivity of restriction enzyme as shown in the table is separated from the cell of the resultant strain. |
