Alpha-L-aspartyl L-phenyl alanine methyl ester dipeptide, for use as a sweetener, is made by forming the peptide bond between the carboxyl gp. of L-aspartine and the amino gp. of L-phenyl alanine methyl ester, after protecting the amino and acid gps. of the L-aspartine, by (a) addn. of dicyclohexyl carbodiimide which reacts with the free carboxyl gp. responsible for forming the peptide bond; (b) activating it by forming an o-acylated deriv.; (c) reacting this without isolation, with the amino gp. of L-phenyl alanine to form the peptide bond; (d) removing the protective gps. from the prod. by hydrogenolysis in water/acetic acid medium in presence of Pd/C catalyst at ambient temp. and atmos. pressure. |
