NEW MATERIAL:A compound expressed by formula I (R1 and R2 are alkyl; R3 is amino or lower alkylamino; R4 is H, alkyl, phenyl or amino) and a salt thereof. EXAMPLE:5-Amino-1,3-dimethylpyrimido[4,5-d]pyrimidine-2,4-dione. USE:A treating and preventing agent for various allergic diseases, e.g. bronchial asthma, uriticaria, allergic rhinitis, allergic dermatopathy or allergic conjunctivitis. PREPARATION:A uracil derivative expressed by formula II (Y is amino, halogen or dilower alkylaminomethyleneamino), e.g. 6-amino-5-cyano-1,3-dimethyluracil, is reacted with a compound expressed by the formula H2N-Z [Z is formula III (X is O or NH), lower alkyl or OH], and the resultant compound is further reduced when the group Z is OH group.
