PURPOSE:To enhance analytical accuracy and sensitivity, by immobilizing an antibody being a substance to be examined and preliminarily reacting a lyposome immunological assaying reagent having a marker substance sealed therein with the specimen containing the substance to be examined before acting the second antibody and complement to the substance to be examined. CONSTITUTION:At least a part of the antibody to a substance to be examined is immobilized on a lyposome receiving lytic action by the activity of a complement without lowering the activity of the lyposome and a marker substance is further sealed in the lyposome to prepare a lyposome immunological assaying reagent which is, in turn, preliminarily reacted with the specimen containing the substance to be examined for a definite time and, thereafter, the second antibody and complement to the substance to be examined are acted. As mentioned above, because the specimen containing the substance to be examined is sufficiently reacted with the lyposome immunological assaying reagent before the second antibody and complement to the substance to be examined are further acted, the accuracy and sensitivity in the quantitative analysis of the substance to be examined can be enhanced to a large extent.
