D-N-Carbamoyl-???-amino acids are prepared by subjecting 5-substituted hydantoins to the action of a cultured broth, cells or treated cells of a micro- organism or an enzyme extracted from the cells, said microorganism being one cultured in a culture medium containing metabolites of pyrimidine bases or their analogs as inducers and having an ability of hydrolyzing the 5-substituted hydantoins so as to substantially produce D-N-carbamoyl-???-amino acids. The accumulation of the enzyme in cells can be further increased, when a metal ion is used in combination with the inducer in the culture. The D-N- carbamoyl-???-amino acids are converted into D-???-amino acids by decarbamoylation using nitrous acid or a microbial enzyme.
