PURPOSE:A preliminarily purified solution containing t-RNA is brought into contact with porous glass bearing benzoyl and diethylaminoethyl groups to adsorb the t-RNA, then it is eluted whereby high-efficiency isolation of the titled t-RNA becomes possible without clogging even under high flow rate operations. CONSTITUTION:Finely ground bovine liver is mixed with tris-hydrochloride buffer solution and with phenol. The mixture is centrifuged and the supernatant is separated and stood after combination with isopropanol. The product is centrifuged to remove the precipitate, the pricipitate is dissolved in tris- hydrocloride buffer solution to remove aminoacid, further treated with ethylene glycol monomethyl ether to exclude glycogen whereby preliminarily purified t-RNA is obtained. The t-RNA solution is passed through a column filled with porous glass bearing benzoyl groups and diethylaminoethyl groups and the adsorbate is eluted to isolate t-RNA specific to aminoacids.
