PURPOSE:To assay the activity of a substrate or an enzyme, by converting hydrogen peroxide generated from an enzyme reaction product due to the substrate or the enzyme in an organism sample into fluorescent substance to which diester oxalate compounds and hydrogen peroxide are added. CONSTITUTION:An enzyme is made to act upon a substrate as component of a living organism or a substrate is made to act upon an enzyme and finally an oxidation enzyme is applied via a single or multiple enzyme reaction processes to form hydrogen peroxide. Then, an non-fluorescent substance and an oxidation catalysis are made to act upon hydrogen peroxide to form a fluorescent substance. Then, diester oxalate and hydrogen peroxide are made to act upon the fluorescent substance to generate light and the intensity of light is measured.
