PURPOSE:To simply measure isolated type thyroxin in blood, by labelling a bond of thyroxin and polypeptide with <125>I or the like to perform a radiation immunoassay using the labelled antigen obtained. CONSTITUTION:A T4-polypeptide bond is obtained from thyroxin (T4) and polypeptide by a glutaric aldehyde method. The bond thus obtained is inactive to a thyroxin-bonded globulin (TBG) and albumin but makes a bond reaction with T4. Then, the bond is labelled with, for example, <125>I or the like to make a labelled antigen, which is used to measure thyroxin T4 in blood by a radiation immunoassay method. Here, for example, a bond of thyroxin and sex gland stimulating hormone labelled with <125>I is used. Thus, the use of the labelled antigen enables sensitive measurement of isolated type thyroxin without being affected by coexisting components in blood, TBG and the like this method is highly effective for the decision on the function of a thyroid gland.
