PURPOSE:A recombinant plasmid for mass producing previously a gene of human growth hormone pre 20K with Escherichia coli, comprising Escherichia coli pBR322, SV40DNA and a gene of human growth hormone pre 20K. CONSTITUTION:A recombinant plasmid for mass producing previously a gene of human growth hormone pre 20K with Escherichia coli, in order to produce human growth hormone 20K with Escherichia coli, etc. This plasmid has a ring-shaped double chain DNA having a structure shown by the fig., and consists of three of Escherichia coli plasmid pBR322, SV40DNA, and a gene of human growth hormone pre 20K (complementary DNA). The plasmid contains ampicillin resistant gene (ampicillin decomposition enzyme gene) derived from plasmid pBR322. Therefore, only Escherichia coli having incorporated the recombinant plasmid can be grown in a medium containing antibiotic ampicillin. A large amount of the gene of human growth hormone 20K can be amplified by cultivating Escherichia coli in the medium. |
