Mature haman leukocyte interferons were prepd. by recombinant DNA technol. using Escherichia coli. Thus, mRNAs for interferons A,B,C,D,F,H, and J were isolated from leukocytes and used as substrates for the enzymic synthesis of ds cDNA. These cDNA were inserted, along with a trp control element, into pBR322 and the recombinant plasmid used to transform E, coli K12 strain 294. Interferon polypeptides have no associated glycosyl residues but is may be modified by an N-terminal methionine or a cleavable conjugate or microbial signal protein at the N-terminus. Interferon known for treament of viral infections and malignancies.
