PURPOSE:To obtain a stable reagent soln. without using an acid and to determine quantitatively bilirubin with good accuracy by subjecting the dye which is formed by bringing a specific diazonium compd. and the bilirubin in a sample into reaction to the measurement of the absorbancy thereof of light having the max. absorption wavelength. CONSTITUTION:The reagent is prepd. by making use of the stable preservability of the diazonium compd. expressed by the formula (X is a halogen) as an aq. soln. and incorporating 0.01-2.0 mg/l such compd. into a buffer soln. such as phosphoric acid, tris-hydrochloric acid or acetic acid having 1-11 pH. Such reagent if added to the bilirubin-contg. bodily fluid (serum, etc) and is allowed to react for 2-20 minutes at 5-50 deg.C. The max. absorption wavelength 535 nm of the dye formed after the reaction is measured. The quantitative determination of the bilirubin in the sample with high sensitivity and accuracy is made possible by using the calibration curve formed preliminarily by using a standard liquid contg. a known ration of bilirubin. The index for the function of the liver is thus obtd. |
