PURPOSE:To provide the titled microorganism containing a plasmid having a specific restriction enzyme incision map, and useful in recombinant DNA technique. CONSTITUTION:A plasmid pBR-LEU having leucine as a nutrient marker is produced from known plasmid pBR325, and is digested with restriction enzyme BamHT. The digested product is added with a desired DNA fragment and linked together by using a ligase, and the resultant linked DNA is used for the transformation of Escherichia coil C600 r-m-. Saccharomyces cerevisiae SHY3 is transformed by the obtained hybrid DNA to obtain the plasmid pLEU135 shown in the Figure, and characterized by a molecular weight of 14.9 kbp, a specific restriction ezyme incision map of a DNA replication starting site (ARA), the number of the restriction enzyme incision sites of pLEU135DNA the molecular weight of the fragment of pLEU135DNA digested with restriction enzyme etc. A transformed strain of Escherichia coil is produced by carrying out the transformation using said plasmid. |
