PURPOSE:To convert signal stranded DNA into technically easily treatable duplex vector by gene recombination operation, by forming cyclic duplex DNA having a specific base sequence of a specific virus. CONSTITUTION:DNA(DNA-a) is obtained by duplexing at least partially single stranded DNA having a length from MYMA of 2637 bases, and DNA(DNA-b) by duplexing at least partially single stranded DNA having a length from MYMV of 2715 bases. The DNA-a and DNA-b are decomposed with a restricted enzyme, and hybrid DNA obtained by integration with a vector for another organism cleaved with the same restricted enzyme, to give hybrid DNA. This hybrid DNA is returned to a host of the original vector and multiplied, to give hybrid DNA. This hybrid DNA is decomposed with a restricted enzyme and bonded through ring closure, to give DNA showing a base sequence. |
