PURPOSE:To enable the essaying of diisopropylaminedichloroacetate (DADA) quickly at a high accuracy, by extracting a nitroso compound generated in the reaction between a sample containing DADA and NaNO2 under an acid condition to apply the extraction liquid to a chromatograph. CONSTITUTION:An NaNO2 aqueous solution is added to an acid aqueous solution of a sample of serum containing DADA having liver strengthening and detoxifying action or the like to react at 60 deg.C for about 10min. Then, the solution is iced and neutralized by a KOH aqueous solution. After the addition of NaCl, hexane is added to the solution, the mixture is stirred up thoroughly and undergoes a centrifugal separation. A fixed amount of a supernatant hexane solution is sampled, benzyl alcohol is added thereto and then the solution adjusted to a fixed capacity correctly by hexane is applied to a normal-phase chromatograph. Or it is subjected to a opposite-phase chromatograph using neutral or polar elution liquid. A quick and highly accurate quantitative analysis of DADA in a DADA medicine or the like can be done by use of a calibration curve obtained based on a known amount of DADA.
